Yeast Two Hybrid cDNA Screening of Arabidopsis thaliana for SETH4 Protein Interaction

SETH4 coding sequence with 2013 bp is a member of gene family expressed in gametophytic tissues of Arabidopsis thaliana. This fragment was PCR amplified using Kod Hi Fi DNA polymerase enzyme. This fragment was cloned into pGBKT7 bate vector and transformed E. coli DH5? cells containing vector were selected on LB medium containing Kanamycin. Finally, pGBKT7-SETH4 bate was transformed into yeast strain Y187 and were selected on SD ?Leu ?Trp medium. After mating of this strain with yeast strain AH109 containing Arabidopsis thaliana cDNA library as prey vector, diploid cells were selected on SD ?His ?Leu ?Trp medium. Screening of cDNA inserted into diploid cells indicated that, 35 colonies out of 240 showing X-gal staining. PCR amplified of cDNA insertion fragments in diploid cells showed some colonies have a chance for SETH4 protein interaction.

Herpes simplex virus type-1 (HSV-1) is a neurotropic pathogen of humans that establishes latent infection in the sensory ganglia innervating the site of primary infection. A number of genes control HSV-1 pathogenicity and latency. Open reading frame P (ORF P) is one of these genes that might have a role in latency and pathogenesis. A complication in the analysis of the role of ORF P in the HSV-1 life-cycle is an antisense overlapping gene, ICP34.5. ORF P is also deleted in ICP34.5 negative mutants and to date, no definite function is attributed to it. An approach to analyze the function of a viral gene such as ORF P is to determine if this gene interacts with any of the cellular and viral proteins both in vitro and in vivo. Therefore, in this work, using GST pulldown assay and Western-blotting, it was investigated that with which cellular and viral gene products ORF P interacts. Our results showed that ORF P interacted with Thymidine kinase (TK) and also with an unidentified cellular protein. Conclusively the results of these works to-gether suggest possible role for ORF P in both splicing and replication of HSV-1.

Transforming growth factor betas (TGF?s) are one of the most widespread and versatile cytokines. The three mammalian TGF? isoforms, ?1, ?2, and ?3, and their receptors regulate proliferation of neuronal precursors as well as survival and differentiation in neurons of developing and adult nervous system. Functions of TGF?s has a wide spectrum ranging from regulating cell proliferation and differentiation, production of extracellular matrix components, chemotaxis, and immunosuppression, to the regulation of cell death. While there is a complete lack or low levels of TGF?1 in the unlesioned nervous system, a widespread expression of TGF ?2 and TGF ?3 can be seen in different areas of the CNS and PNS. Consistent co-expression of TGF?2 and ?3 in neurons, astroglial, and Schwann cells is indicative of their multiple effects on neurons and glial cells. Based on overlapping expression of TGF ?2 and TGF ?3 and therefore their functions, our study was designed to differentiate between the functions of the two isoforms during development. Because mice lacking both isoforms die around embryonic day 15 (E15), we decided to use single homozygous and double heterozygote null mutant embryos who survive and lack any abnormal nervous system phenotype after birth. The expression pattern of markers for extracellular matrix protein such as reelin and chondroitin sulfate proteoglycan (CSPG) as well as those for cytoskeletal proteins such as microtubule associated protein (MAP), a marker for differentiated neurons, was studied in E14.5 and E16.5 mice embryos both by immunohistochemistry and in situ hybridization. Despite a reduced expression of CSPG and MAP proteins that was seen only in TGF beta 3 null mutants, Tgf?2+/+Tgf?3?/?, no abnormal localization of these markers and no change in the expression at mRNA level was noticed. The normal localization of the markers as well as their unchanged mRNA expression suggest that TGF ?2 compensates for the lack of TGF ?3 and therefore the two isoforms act in parallel to ensure the stability of brain cytoarchitecture during development.

Spectrophotometric Simultaneous Kinetic Determination of Iodide and Iodate Using Partial Least-Squares Calibration Method in a Single Kinetic Run

A rapid, sensitive and versatile kinetic method is presented for the simultaneous spectrophotometric determination of iodide and iodate by partial least-squares regression (PLS) using original and derivate data named as absorbance and rate data. The method is based on the catalytic effect of the cited anions on the reaction rate between Ce(IV) and As(III) in 2 mol l?1 sulfuric acid medium. The Savitzky-Golay convolution method is used for calculating and smoothing the rate data. Results show that PLS is an excellent calibration method to resolve the mixtures of two anions by first-order or pseudo first-order kinetic procedures without any previous knowledge about rate constant values. The 26 calibration solutions were made of iodide and iodate in the range of 10-48 and 55-235 ng ml?1. The RMSEP calculated for 12 test solutions are 0.1756, 0.5074 and 0.2327, 0.9197 for the absorbance and rate data for iodide and iodate respectively. The application of the method was confirmed by analysis of these anions in real matrix samples.

Synthesis, Characterization and Studies on Catalytic Behavior of Mn(?) Complex with 2, 2? Bipyridine, 1, 1? Dioxide Ligand within Nanoreactors of MCM-41

Manganese (?) complex with 2, 2? bipyridine, 1, 1? dioxide (bpdo) ligand was immobilized within nanoreactors of MCM-41. The immobilized complex was characterized by powder x-ray diffraction (XRD), nitrogen adsorption desorption, and FTIR. The pore volume, surface area and pore diameter of MCM-41 decreased after immobilization of Mn complex. Three bands observed at 1250, 1220 cm?1 and 757 cm?1 are attributed to the NO stretching and bending vibrations respectively. It was found that the immobilized Mn complex on MCM-41 successfully catalyzed the oxidation of cyclohexene, styrene, norbornene, and trans-stilbene with tert-butylhydroperoxide (TBHP).With the exception of cyclo-hexene, the other three olefins afforded the corresponding epoxides efficiently. Preparation and catalytic effect of Mn salt either on MCM-41 or modified MCM-41 with aminopropyltriethoxysilane; MCM-41(AP) will be discussed.

Middle Devonian-Early Carboniferous Conodont Faunas from the Khoshyeilagh Formation, Alborz Mountains, North Iran

Conodonts of Khoshyeilagh Formation, the most complete section of Devonian rocks in the Alborz Mountains, northern Iran, has been studied, and subspecies of the genera Icriodus, Bipennatus, Polygnathus, Pelekysgnathus, Ancyrodella, Palmatolepis, Bispathodus, Scaphignathus, Clydagnathus, Gnathodus, Scaliognathus and Dollymae have been obtained from the studied section. The faunas indicate an Eifelian to Tournaisian age. The falsiovalis, Late rhomboidea-Early marginifera, Latest marginifera-trachytera, Middle expansa-Middle praesulcata, bouckaerti and anchoralis-latus conodont zones have been identified in the formation. Six new species are described: Icriodus ariaei, Ic. alborzensis, Ic. khoshyeilaghensis, Polygnathus klapperianus, Pelekysgnathus housei and Pe. talenti.

Arens Regularity and Weak Amenability of Certain Matrix Algebras

Motivated by an Arens regularity problem, we introduce the concepts of matrix Banach space and matrix Banach algebra. The notion of matrix normed space in the sense of Ruan is a special case of our matrix normed system. A matrix Banach algebra is a matrix Banach space with a completely contractive multiplication. We study the structure of matrix Banach spaces and matrix Banach algebras. Then we investigate Arens regularity and weak amenability of certain matrix algebras which are built on matrix Banach algebras. In particular we show that for such algebras both of Arens regularity and weak amenability problems can be reduced to the same problem for a considerably smaller algebra.

We propose a method of estimation of the derivatives of probability density based on wavelets methods for a sequence of random variables with a common one-dimensional probability density function and obtain an upper bound on Lp-losses for such estimators. We suppose that the process is strongly mixing and we show that the rate of convergence essentially depends on the behavior of a special quadratic characteristic.
